چکیده
Acyl-CoA: cholesterol acyltransferase (ACAT, EC 2.3.1.26) is responsible for intracellular esterification of cholesterol and plays a key role in intestinal absorption of cholesterol, hepatic production of lipoproteins1,2) and accumulation of cholesteryl esters within macrophages and smooth muscle cells of the atheroma3). Therefore, ACAT is an attractive target for new treatments of hypercholesterolemia and atherosclerosis4). In the course of our search for ACAT inhibitors from microbial sources, phenylpyropene C was isolated from the fermentation broth of Penicillium griseofulvum F1959. It was deposited in the Korean Collection for Type Culture (KCTC), Korea, as KCTC 0387BP. The fungal strain F1959 was originally isolated from a soil sample collected at Ulsan, Korea. Taxonomic studies of the strain were carried out by the method of Samson5). Czapek's medium (0.2% NaNO3, 0.1% K2HPO4, 0.05%

Acyl-CoA: cholesterol acyltransferase (ACAT, EC 2.3.1.26) is the primary enzyme responsible for intracellular esterification of cholesterol in intestinal mucosal cells, synthesis of the cholesteryl esters packaged into very-lowdensity lipoproteins (VLDL) secreted by liver, accumulation of cholesteryl esters within macrophages and smooth muscle cells of the atheroma, and plays a key role in form...
متن کاملPenicillium griseofulvum F1959, high-production strain of pyripyropene a, specific inhibitor of acyl-CoA: cholesterol acyltransferase 2.
Acyl-coenzyme A: cholesterol acyltransferase (ACAT) catalyzes cholesterol esterification and plays an important role in the intestinal absorption of cholesterol, hepatic production of lipoproteins, and accumulation of cholesteryl ester within cells. During the course of screening to find ACAT inhibitors from microbial sources, the present authors isolated pyripyropene A from Penicillium griseof...
متن کاملIsochromophilones III-VI, inhibitors of acyl-CoA:cholesterol acyltransferase produced by Penicillium multicolor FO-3216.
New azaphilones named isochromophilones III-VI were isolated from the culture broth of Penicillium multicolor FO-3216 as inhibitors of acyl-CoA: cholesterol acyltransferase (ACAT). Their structures were elucidated by NMR and other spectroscopic analyses. The IC50 values of isochromophilones III, IV, V and VI for ACAT activity in an enzyme assay using rat liver microsomes were calculated to be 1...

Penicillium purpurogenum FO-608, a soil isolate, was found to produce a series of new inhibitors of acyl-CoA:cholesterol acyltransferase (ACAT). Three active compounds, designated purpactins, A, B and C, were isolated from the fermentation broth of the producing strain by solvent extraction, silica gel column chromatography and HPLC. Purpactins inhibit ACAT activity in an enzyme assay system us...

A new inhibitor of acyl-CoA:cholesterol acyltransferase (ACAT), designated lateritin, was isolated from the mycelial cake of Gibberella lateritium IFO 7188 by successive purification procedure of solvent extraction, silica gel column chromatography and reverse phase HPLC. Spectroscopic analyses of the compound yielded 4-methyl-6-(1-methylethyl)-3-phenylmethyl-1,4-perhydrooxazine-2,5- dione as t...
